fa ارزیابی ارتباط پلی مورفیسم تک نوکلیوتیدی (rs2275913) در ژن اینترلوکین 17 با عفونت مزمن هپاتیت B در مراجعه کنندگان بیمارستان طالقانی تهران ویروس شناسی Virology مورد-شاهدی Case-Control <div style="text-align: justify;"><span style="font-family:times new roman;"><strong><span style="color:blue;"><span style="font-size:10.0pt;">سابقه و هدف:</span></span></strong> <span style="font-size:10.0pt;">بیماری هپاتیت </span><span dir="LTR"><span style="font-size:10.0pt;">B</span></span><span style="font-size:10.0pt;"> یکی از دلایل&nbsp; اصلی در التهاب و آسیب های کبدی است که می تواند منجر به عفونت مزمن ویروسی هپاتیت </span><span dir="LTR"><span style="font-size:10.0pt;">B</span></span><span style="font-size:10.0pt;"> شود. پلی‌مورفیسم تک نوکلئوتیدی در ژن سایتوکاین ها می تواند بر پاسخ ایمنی میزبان تاثیر بگذارد. اینترلوکین ۱۷ که توسط سلول های </span><span dir="LTR"><span style="font-size:10.0pt;">Thelper17</span></span><span style="font-size:10.0pt;"> تولید می شود مشخص شده که در عملکرد سیستم ایمنی در بیماری های عفونی و التهابی نقش باز‌ی می کند. این مطالعه به منظور بررسی ارتباط میان پلی مورفیسم در ژن</span></span><span style="font-family:times new roman,serif;"><span style="font-size:10.0pt;">rs</span></span><span style="font-family:times new roman,serif;"><span style="font-size:10.0pt;">2275913) </span></span><span style="font-family:times new roman,serif;"><span style="font-size:10.0pt;">IL-17)</span></span><span style="font-family:times new roman;"><span style="font-size:10.0pt;">&nbsp;و عفونت مزمن هپاتیت </span><span dir="LTR"><span style="font-size:10.0pt;">B</span></span><span style="font-size:10.0pt;"> انجام شد.</span> <span style="font-size:10.0pt;"></span><br> <strong><span style="color:blue;">مواد و روش&shy;ها:</span></strong> این مطالعه موردی-شاهدی بر روی ۱۳۰ بیمار مزمن و ۱۳۰ فرد سالم به عنوان گروه شاهد انجام شد. افراد بیمار با نتیجه مثبت تست الایزا برای <span dir="LTR">HBsAg</span> و <span dir="LTR">Anti-HBc Ab</span> و افراد شاهد با نتیجه منفی این تست وارد مطالعه شدند. برای تعیین ژنوتیپ در هر دو گروه از <span dir="LTR">DNA</span> استخراج شده از نمونه خون با روش <span dir="LTR">Salting out</span> استفاده شد. تعیین ژنوتیپ با روش <span dir="LTR">PCR-RFLP</span> انجام گرفت. <span dir="LTR"></span><br> <strong><span style="color:blue;"><span style="font-size:10.0pt;">یافته ها:</span></span></strong> <span style="font-size:10.0pt;">فراوانی ژنوتیپ در </span><span dir="LTR"><span style="font-size:10.0pt;">rs</span></span><span dir="LTR"><span style="font-size:10.0pt;">2275913</span></span><span style="font-size:10.0pt;"> تفاوت معنی داری بین دو گروه بیمار و شاهد نشان نداد. توریع ژنوتیپ در گروه بیمار به صورت </span><span dir="LTR"><span style="font-size:10.0pt;">GG</span></span><span style="font-size:10.0pt;"> 40/8%، </span><span dir="LTR"><span style="font-size:10.0pt;">AG</span></span><span style="font-size:10.0pt;"> 41/5% و </span><span dir="LTR"><span style="font-size:10.0pt;">AA</span></span><span style="font-size:10.0pt;"> 17/7% و در گروه شاهد </span><span dir="LTR"><span style="font-size:10.0pt;">GG</span></span><span style="font-size:10.0pt;"> 42/3% و</span><span dir="LTR"><span style="font-size:10.0pt;">AG</span></span><span style="font-size:10.0pt;"> 45/4% و </span><span dir="LTR"><span style="font-size:10.0pt;">AA</span></span><span style="font-size:10.0pt;"> 12/3% مشخص شد (0/469</span><span dir="LTR"><span style="font-size:10.0pt;">p=</span></span><span style="font-size:10.0pt;">).</span><span style="font-size:10.0pt;"></span><br> <strong><span dir="RTL"><span style="color:blue;"><span style="font-size:10.0pt;">نتیجه گیری:</span></span></span></strong> <span dir="RTL"><span style="font-size:10.0pt;">نتایج مطالعه ارتباطی میان پلی مورفیسم در ژن </span></span><span style="font-size:10.0pt;">IL-17 rs2275913</span><span dir="RTL"><span style="font-size:10.0pt;"> با بیماری هپاتیت </span></span><span style="font-size:10.0pt;">B</span><span dir="RTL"><span style="font-size:10.0pt;"> مزمن نشان نداد</span></span></span></div> <div style="text-align: justify;"><span style="font-family:times new roman;"><span style="color:#0000FF;"><strong>BACKGROUND AND OBJECTIVE:</strong></span> Hepatitis B disease is one of the main causes of inflammation and liver damage that can lead to chronic hepatitis B virus infection. Single nucleotide polymorphism in the cytokines gene can affect the host immune response. Interleukin 17 produced by Thelper17 cells has been shown to play a role in immune function in infectious and inflammatory diseases. This study was conducted to investigate the association between polymorphism in IL-17 gene (rs2275913) and chronic hepatitis B infection.<span dir="RTL"></span><br> <span style="color:#0000FF;"><strong>METHODS:</strong></span> This case-control study was performed on 130 chronic patients as a case group and 130 healthy individuals&nbsp; as control. Patients with positive result of ELISA test for HBsAg and Anti-HBc Ab and control subjects with negative result of this test were enrolled. PCR-RFLP was used to genotype extracted DNA from blood samples.<br> <span style="color:#0000FF;"><strong>FINDINGS:</strong> </span>The genotype frequencies of rs2275913 did not show significant difference between patients and control groups.
